The story revolves around the title character, Karin, the middle child in a family of vampires who immigrated to Japan two centuries earlier. However, Karin is different from the rest of the family in that she produces blood instead of feeding on it; because of this, she has to inject blood by biting someone or else she suffers exaggerated nosebleeds. (In manga and anime nosebleeds are also a metaphor for sexual attraction; as Karin is a coming of age story dealing with her first love, the nosebleed imagery does double-duty.) Because of her unusual condition, Karin lives her life just as an ordinary teenage girl would, despite being considered something of a freak by her family. She is able to venture outside during the day and attend high school, and follows the sleep pattern of normal humans. However, the general peacefulness of her life is disrupted with the arrival of a new transfer student, Kenta Usui.

Any time she goes near him her blood increases. At first she tries to avoid him but it doesn't work since they are in the same class and work in the same restaurant. Kenta on his part thinks that there is something fishy about Karin and eventually he learns her secret. Karin's elder brother Ren tries to erase his memory but Anju, Karin's younger sister stops him from doing so. She also convinces their parents Henry and Carrera to make Kenta their ally, because he can help Karin during the daylight.

Karin and Kenta become friends. Kenta helps Karin clean up the mess made by her nosebleeds and Karin prepares him a lunch box (bento) every day.

At this point the manga and anime diverge. In the anime only, another transfer student appears, Winner Sinclair. He is very good looking and most of the schoolgirls fall for him including Karin's best friend Maki Tokito. But Winner states that Karin is the girl of his dreams. He also is a vampire hunter much to Karin's dismay and Kenta's annoyance because he receives a duel challenge over Karin almost every day.

As the story progresses Kenta and Karin's attachment to one another grows, and so do their problems. First Karin's grandmother Elda breaks free from her coffin (which Carrera had sealed her into) and starts wrecking everyone's lives. Except for her bust size, hair length, and hair color she looks identical to Karin, and uses this resemblance to go unnoticed in Karin's school.

She attempts to drink Kenta's blood, because she believes humans and vampires cannot coexist, due to her romance to Alfred Sinclair which ended in tragedy. Eventually, she gives up and returns to her casket, though returns quickly as she apparently found out how to convert Karin to a full vampire. Shortly after, Anju's powers also awake and Victor Sinclair(Winner's grandfather) arrives to hunt the vampires down.

In Karin, vampires (kyūketsuki) are somewhat like traditional vampires of folklore and horror films, however there are also significant differences. Like traditional vampires of folklore they burn up in the sunlight, can control familiars, as well as possessing powers to pin humans down and erase their memories. However, young vampires whose blood-sucking instincts have not yet manifested can withstand subdued daylight and eat human food, but this changes when they hit a certain stage in their development, the vampire equivalent of puberty. From this point on, they become heliophobic and lose their sense of taste for human foods; Ren Maaka described the lunch Karin made for him as tasting like sand.

Karin differs from much vampire literature in that it depicts vampires as not being evil as such, and in fact generally not being interested in humans other than as a source of nutrition.[5] Rather, they are shown almost as victims, their main concern being to remain unnoticed so as to avoid being persecuted (and executed) by humans.

Another difference is that each vampire has his or her own taste in blood based on the personality trait of the human beings on whom they feed. When they suck that person's blood, the associated personality trait is also drained. For example, Karin's taste is misfortune (不幸, fukō?), so she is drawn to misfortunate people. On biting such a person, that person will lose all their sorrow and become happy, although the effect wears off over time. So in Karin, vampires are able to have a positive effect on humans, but this effect is not intentional[6] and depends on the vampire having a taste for a negative trait. However where their taste is for something positive such as love (as is the case with Karin's grandmother, Elda), the effect of the vampire on human society can be devastating. Since humans and vampires are two completely different species, being bitten by a vampire does not cause a human to become a vampire.

In Karin, the countermeasures against vampires mentioned in vampire mythology are depicted as mostly ineffectual, little more than urban legends. For instance, Karin states that while garlic possesses no magical powers against vampires, it is a combination of the vampires' superhumanly acute sense of smell and the garlic's odor that makes vampires shun it. Similarly, a wooden stake through the heart is not necessarily fatal - both Elda and Henri Marker have survived such - as well as being shot (Ren), stabbed (Elda), run through (Elda and Carrera), skull broken by pistol-whipping (Anju), or even severe burning from prolonged exposure to sunlight (Henri). Their bodies enter a death-like state consistent with the nature of their "fatal" injuries, in which they regenerate completely with no visible scarring, although they will be rather hungry afterwards. Nevertheless, in the manga, vampires as a species are dying out. Those who escaped from the mainland have a low birth rate (zero in the year of the story) while several passed away; the Marker family are the only vampires to have produced children in a century.

In terms of appearance, apart from a few distinguishing features vampires appear to look like normal humans (manga/anime hair and eye color nonwithstanding). The first and foremost non-human feature would be their fangs; under normal situations they appear as nothing more than pronounced canine teeth, but when hungry (or angry) they extend to carotid artery penetration length. Their eyes also appear normal until they are excited, when their round pupils turn into cat-like vertical slits. They also have claw-like fingernails.

Another noticeable feature is their low body temperature due their being nocturnal. As part of their heliophobia, along with their eyes having excellent vision at night yet sensitive to light (both natural and artificial), their bodies have evolved to survive cooler nighttime temperatures. As a result, they tend to overheat if they are out during the day and are at risk of severe sunburn, which could lead to actual burning. In episode 16 of the anime, vampires are executed by prolonged exposure to sunlight, incinerating their bodies into charcoal and ash.

Karin is a comedy which makes use of many devices, the central one being embarrassment: indeed the chapters of the manga are enumerated as her many embarrassments or shames (恥, haji?). For the demure and somewhat shy Karin, the world is full of embarrassments, not least of which is being not just a vampire but a defective one.

The traditional sexual overtones of vampirism are also a source of comedy. One example of this is when the sight of Karin seemingly necking people is initially misinterpreted by Kenta as Enjo kōsai (in other words, he thinks she's a teenage prostitute). As a rather demure girl, Karin is mortified by the misunderstanding, but unable to correct it because to reveal the truth would be even worse. Another example is Karin's attraction to Kenta which provides an opportunity for comedic confusion between love and blood lust, symptoms such as a pounding heart being common to both. Meanwhile her brother Ren clearly enjoys sexual relations with his victims, and his overt displays of lust are another source of embarrassment for the much more reserved Karin.

Blood is consistently used for comic effect rather than horror. Karin's profuse nosebleeds (鼻血, hanaji?) serve both as a pun on the common manga symbolism of nosebleeds as indicating sexual arousal (the latter causing an increase of blood pressure; the prior can be caused by an increase of blood pressure), and as a source of menstruation jokes: indeed the author originally envisaged Karin "bleeding from every orifice" until it was vetoed by her editor.[1] In the anime, a standing joke is that the vampire hunter Winner Sinclair, far from being a hardened killer, actually faints at the sight of blood, such as every time Karin has a nosebleed.

Karin's buxom physique (she officially possesses an E-cup) is a source of ecchi (lewd) humour, especially when it inspires jealousy in other females, such as Maki and her grandmother (the latter an A-cup). In the anime, a ritual supposedly to cure her of her blood-producing condition turns out to actually be a cure for increasing bust size, and the sight of her exposed breasts afterwards proves enough to cause a vampire hunter to faint, thus defeating him.

Karin Maaka (真紅 果林, Maaka Karin?)

The protagonist of the show. Karin is a 16-year-old "blood-producing" vampire (in Japanese, 増血鬼 zōketsuki — literally "haematopoietic demon"; rather than needing to acquire blood from humans, her body produces too much blood instead (polycythemia). As such, she does not suck blood from her "victims"; rather, she injects her surplus blood into them. This has the effect of making her victims very lively and positive. To make matters worse, her blood levels increase dramatically whenever she senses unhappiness (fuk&#333 in those around her, or if she gets embarrassed about something. This results in frequent and excessively overwhelming nosebleeds, followed by anaemic symptoms such as light-headedness, with cause frequent visits to the school nurse's office so she can rest.
Unlike the rest of her family, Karin is a daywalker who can stand under the Sun without getting scorched. She attends the local high school and works part-time at Julian's Restaurant. She is generally has a cheerful demeanor but tends to worry about things and is easily embarrassed. She is good with children and enjoys cooking. Her hair is purple. Though Winner considers her to be the ideal woman, she is not particularly popular with boys; when she was younger, she was teased by boys at her school who referred to her as "Nosebleed Girl".
Kenta Usui (雨水 健太, Usui Kenta?)

Kenta Usui is a student that transferred to Karin's high school. He is also her workmate. Kenta is the only human that knows of Karin's vampiric nature. He learned what she was after witnessing her biting three victims, one of which was his own mother. He has promised to keep this knowledge a secret. As his unhappiness causes Karin's blood to rapidly increase, she has taken it upon herself to cheer him up by making him a bento every day. He could not afford to buy his own lunch and is therefore grateful to her.
Karin's parents did not approve of Kenta's knowledge of Karin's secret at the beginning until it occurred to them that he would be a strong ally as he could watch over Karin during the daytime. Kenta ends up falling in love with Karin, though he does not show it clearly until Episode 13 onwards (or when he confesses).
He lives with his mother, Fumio Usui (雨水 文緒, Usui Fumio?). His father, Shusei Iizuka (飯塚 修成, Iizuka Shūsei?), left them, though as a child he was told that his father was dead. He tries hard to take care of his mother in his father's place and overworks himself at the restaurant to earn money for her.
He has an unusual appearance; the pupils and irises of his eyes are very small and his neutral expressions are often mistaken for scowls. Early on in the story, his school colleagues commented upon his strange facial appearance, but they accepted him in later episodes. His face, hair-style, and mannerisms are quite similar to those of Ichigo Kurosaki from Bleach.
Anju Maaka (真紅 杏樹, Maaka Anju?)

Karin's little sister, very much in the Gothic Lolita mold. She has long silver hair and amber eyes and is often seen carrying Boogie, or "Boogie-kun" as she refers to him, a talking, cleaver-wielding, blue-haired doll. Anju is very soft-spoken, and her vampiric nature has not yet been fully awakened at the start of the series. She controls a horde of bats, which watch over Karin during the day, and she also has the power to erase the memories of humans, a power which Karin herself does not have. Overuse of her powers often causes her to pass out. She appears to be perpetually calm and collected, showing very little emotion in her expression or speech, unlike her highly-strung parents. Based on her behavior, she appears to be the most mature and sensible member of the Marker family.
Anju is very attached to her sister and the two of them look after each other. Anju watches over sister with the use of her bats, helps her with feeding by erasing Karin's victims memories and serves as an intermediary between Karin and Kenta. She occasionally becomes jealous of those who are close to Karin. Anju attends fifth grade at an elementary grade school on days that are overcast or rainy. She gets a lot of attention from the boys.
In Episode 20 (near the ending of the series) Anju's vampire instincts awaken, and her preference of blood is identified as jealousy. Karin is horrified when she finds out who her first victim is; Maki Tokitou.
Ren Maaka (真紅 煉, Maaka Ren?)

Karin's handsome blue-haired older brother. He a bit of a lounge lizard with an eye for the ladies (especially those who are stressed, as stress is his blood preference, though, as a self-confessed pervert, he enjoys the other benefits he receives from encounters with his victims besides their blood). He is scornful of Karin's condition and considers her to be almost a non-vampire, yet he is the one who takes her out and realizes that Karin's blood increases are due to the unhappiness of others. Ren usually will only bite women but Kenta, who in addition to being unhappy is very stressed, drives Ren crazy. Ren's only fear is Elda Marker, who nearly killed him when he was four by smothering him with affection.

Henry Marker (ヘンリー・マーカー, Henrī Mākā?)

Karin's, Anju's, and Ren's father, also the husband of Carrera. As a typical loving father, he is very protective of Karin, especially with regards to her relationship with Kenta. When it seems their relationship is becoming more than platonic, his fatherly instinct drives him to intervene, which leads to very embarrassing moments for the couple. In the past he has risked his life to save his daughter, for which she is extremely grateful. His taste in blood is pride and he sucks blood the blood of conceited people.

Carrera Marker (カレラ・マーカー, Karera Mākā?)
Voiced by: Emi Shinohara (Japanese), Chio Su Ping (English)
The Marker family matriarch. She is quite domineering and usually keeps her husband under her thumb. Her preference is the blood of liars, which she drinks like a fine wine. Carrera's only known enemy in the anime is Elda Marker, as discovered in Episode 15. In the English version she is known as Calera Marker.
Boogie-kun (ブギーくん, Bugī-kun?)

Anju's sarcastic talking doll. He often tells Karin how useless she is, which usually leads to Anju punishing him. The other dolls in Anju's room also talk and have their own personalities, but "Boogie-kun" is her favorite. Boogie-Kun is actually a murderers' spirit trapped in a doll who becomes upset when he is away from Anju too long. In Volume 3 of the manga one of Anju's classmates takes Boogie-kun, with grievous consequences until Anju comes to take Boogie-kun back. In the anime, Boogie-kun was Anju's anger in the form of a doll so after Anju awakens as a full vampire and shows her anger towards Boogie-kun and Karin in Episode 20 he reverts back to an inanimate doll. The voice of Boogie-kun in the anime was modelled after the glove puppet Mr.Hat from the American cartoon South Park.[7]
Elda Marker (エルダ・マーカー, Eruda Mākā?)

Karin's grandmother. Karin bears a remarkable resemblance to her; the only difference any of the characters seem to notice is Elda's smaller bust (an A-cup; Karin gets her E-cups from her mother's side), but she can also be told apart from Karin by her much longer pink hair and her red eyes. Her first appearance in the anime is in Episode 15, when she breaks free from her sealed coffin in the Marker family mansion. Elda's blood preference is love. Ren is scared stiff of her; he first met her when he was four, before Karin and Anju were born, and she almost killed him with affection. She seems determined to interfere with the normal flow of the Marker family's life, and goes to Karin's school at night while all the students are working on their culture festival projects. Her affliction is love; she once loved a human being, and the results were disastrous. Elda is unaware of Karin's affliction until events come to a head at the school festival, and Karin suffers a blood surge. Following this, Elda goes in search of a "cure" for the condition - and as of the end of Episode 20, she seemed to have found it, but in the end it turned out to be a cure for "breast-increase" rather than blood-increase.
Maki Tokitou (時任 麻希, Tokitō Maki?)

Karin's best friend at school. She often wears her long hair in a ponytail. Her family owns a book store. She is a cheerful girl who supports Karin through all her troubles, but it doesn't stop her from ribbing Karin mercilessly about Kenta. Maki does not know Karin is a vampire. She was the only girl who didn't swoon over Winner when he first arrived at school, but she is slowly starting to fall for him, despite the fact that he's always trying to romance Karin. Winner always refers to Maki as "Karin's friend", much to Maki's annoyance, and the hunter's blindness to all but Karin led to Maki being shot down in flames when she tried to give him a Christmas present (and confess at the same time). This brought forth feelings of jealousy in her, meaning she was to become Anju's first victim, to Karin’s utter horror.
Fumio Usui (雨水 文緒, Usui Fumio?)
Voiced by: Aya Hisakawa (Japanese), Meriem Bekka (English)
Kenta's mother. She is often very depressed since she has trouble holding a job. In each job she has, many men will fall in love with her. When she refuses them they start to harass her which leads to her getting fired. It was commented by the parents of Karin that she may give off a great deal of pheromones that are attracting men to her. In Episode 2, Karin bites and transfuses blood into Fumio, causing her to be happy and energetic all the time; Kenta sees this and does not know what to think, but he is happy that he can see her smile instead of being depressed.

Shusei Usui (, Shusei Usui?) (Manga only)
Kenta's father. Shusei appears in volume 6 in the manga looking for Kenta and Fumio (along with a detective sent by Fumio’s mother, and is constantly stalked and harassed by Shusei), but Kenta refuses to accept him, saying "You're a stranger" or "I have no father".
Winner Sinclair (ウィナー・シンクレア, Winā Shinkurea?) (Anime Only)

A vampire hunter descended from a long line of hunters. Winner is an anime-only character. He transfers to Karin's high school and falls in love with her at first sight, constantly making romantic overtures toward her. Despite being a vampire hunter, he does not know about Karin's true nature, as he is blinded by love and possibly also because he is not very bright. As a child, he met a girl with whom he shared his first kiss. As a hunter, rejuvenated by the first flush of love, he redoubled his efforts. However, the girl he "kissed" was (unbeknownst to him) actually a young Karin, who bit him and injected her blood into him. The effect of her blood seems to have been permanent in Winner's case; in keeping with the series premise, instead of a vampire siring another vampire, an anti-vampire has sired a vampire hunter — albeit an overly-enthusiastic and largely ineffectual one. He is first seen in Episode 7 of the anime.
Winner is very melodramatic, and his hunting garb is flamboyant: a black, flowing Neo-style long overcoat, with a white chest and a gold "W" plastered across the front, and a huge wide-brimmed hat with a feather sticking out of it. His blond good looks make him extremely popular with the girls at the school and his "cool" appearance is also commented on by passing strangers.
There is one massive stumbling block to Winner's progress as a vampire hunter — he has a fear of blood (hemophobia), even going so far as feeling queasy when seeing spilt red paint. However, he did manage to overcome this dread in one episode when Karin suffered a blood attack, and carried her to safety - so he does have his good points. Also, in Episode 8, the alarm on his phone appeared to be that of the Rocky Balboa theme. Winner appears first in the anime at Episode 7. His looks and personality are similar to Tamaki Suoh From Ouran High School Host Club.
